Replicates
종종 biodata를 수집하다보면, 아래와 같은 두가지 종류의 replicate을 발견한다.
- Technical replicates: a test performed on the same sample multiple times; i.e., if there are triplicate non-treated samples, a technical replicate would be testing sample #1 of the non-treated multiple times
- Biological replicates: a test performed on biologically distinct samples representing an identical time point or treatment dose; i.e., if there are triplicate non-treated samples, a biological replicate would be testing sample #1, 2 and 3 of the non-treated group
Technical replicates는 장비와 같은 기술적인 부분으로 오는 noise를 결정하고 조절하기 위해 최대한 동일한 조건에서 같은 샘플로부터 반복 채집한 자료를 의미한다. Assay나 technique의 reproducibility를 조절하기에 좋다.
반면 Biological replicates는 그룹간의 차이와 같은 생물학적인 차이로부터 오는 정보를 잡기위해 서로 다른 샘플로부터 채집한 자료를 의미한다.
좋은 실험은 일반적으로 biological replicate를 통하여 noise를 조절하고, 생물학적인 차이를 보기위해 technical replicate을 활용하여, 보다 정밀하게 샘플을 정돈하여 분석을 진행한다.
하나의 예시를 들자면, LINCS L1000 data set의 마지막 단계의 자료형인 level 5에서는 replicates 3개 (vector 3개)의 서로에 대한 spearman correlation을 확인한 뒤 정규화하여 가중치를 두어 하나의 샘플 (vector)로 만든다.
